We plan to stay at the Grand Floridian mid September.

Can you specify a specific building, such as Boca Chia?

Does anyone have any specific room numbers to request, I'd like a third or fourth floor room in the center of the building, near the elevator.

sure and being september it will probably be slower. So it will be easier for you to request it. But always remember it is a request.

I had read recently on another thread that:

1. Room assigners don't like to be asked for specific room #'s

2. They'd prefer to be told what you are looking for in a room, so that if they can't get the # you've requested, they know what you like and can get something comparable for you.

Can't vouch for any of this since I'm only repeating what I read, but it makes sense to me. I guess if you tell them the locations and views you'd prefer and why, they'll do their best to make you happy.
