We are making reservations for our family to go to different places to eat. There are 4 adults and 1 infant in our party. If my mother and sister decide not to come to one of our meals will Disney change the number in our party or will we lose the reservation?

As long as it's not somewhere that requires prepayment (CRT, HDDR, SOA, MBBQ), or deposits (CG), you should be able to show up and let them know at that time that 2 were unable to attend. You shouldn't have any issues!;)

You'd have to cancel the reservation and book a new one for the new party size. You can do that if there is availability for the new party size. That's why it's suggested that you just show up and say they couldn't make it, unless of course it is a prepaid meal or one for which you'd be charged a no-show fee.
