LOVE BI! My brother owns a home there so we are lucky to visit often. It is a great daytrip from MA. We usually take 395 south to Exit 85 to 138south past URI to 108 which takes you straight to the ferry.
If you don't mind paying more, the high speed is the way to go. gets you there in 30 minutes.
A couple hotels are the Spring House, the National and the Hotel Manisses. All are within walking distance of town.
I would suggest renting bikes or mopeds (but they can be dangerous). You can pretty much bike all around, but it is hilly. The taxis will also do island tours.
The town has some cute shops. We love going to the Spring House for lunch or cocktails. Great views!
Also..see the Southeast lighthouse...can hike down the stairs to the beach there, but it is not easy coming up.
Beaches...Town Beach has concession stand, showers, etc. Our favorite is Mansion Beach at the end of Mansion Road.
Some other places we enjoy...Dead Eye Dicks for dinner...expensive though.
Champlins Marina has a pier with a bar at the end which is great to watch the boats in the harbor. They also have a place to get snacks and ice cream.
Another more reasonable restaurant is The Oar....anyone you ask will point you there. Good food and reasonable prices.

PS..Ballards on the beach (in town) is fun. They usually have music playing on the beachside. BUT...the food is awful...great for cocktails though.
