Yes you can. Usually I put them on the floor and put one leg through a strap to secure it. On some rides, there are small pouches - if the backpack is squishy enough, it can go in there. On GRR, you can use a locker for free to keep your stuff dry.
I always put my leg through the backpack strap too, mostly because I don't want to forget it. I never use those little pouches on rides- those are just asking you to forget your stuff. If I'm not in charge of the backpack, then I just make sure my purse strap is around me. We do put the backpack in the pouch on Soarin. We have never come across a ride where the backpack didn't fit somewhere.

good to know about the free locker on GRR, on water rides I usually wear a poncho and the bag is on the front of me under the poncho
some of the rides (indiana jones...) have little net things to hold your items, usually i just wrap them around my leg or put them on my lap/under the lap bar. depending on the ride.
I just kept my backpack on my back on most rides...none of the CM seemed to mind and I only had jackets in the bag so it was pretty squishy. Although on Indiana Jones, I was trying to put something in the pouch (it wouldn't fit in my backpack) and the ride started! I was in the front right hand side so I wasn't going to risk it falling out of the side of the car so the person next to me held it between their legs.
