What is the easiest way to get from Magic Kingdom to Pleasure Island at night ? Is there a boat, or is the bus the only way ?

No boat. (if there was, you wouldn't want to take it... that'd be like a 45 minute trip if the lakes were connected.)

From MK, either walk over or take monorail to Contemporary and pick up bus to Downtown Disney from there (or get a cab).

Personally, if you are not already staying there, I'd avoid the Contemporary... the reason being, that bus typically gets crowded enough with Contemporary guests.

What I would suggest, is taking the Old Key West bus. It's never crowded. Get off at the first stop, and then wait for the DTD bus. DTD is only a 5 min ride from OKW, and that bus is also rarely crowded.

Well since you're staying at the Dolphin, isn't there a bus from either there or the BWI,YC/BC that takes you to DTD?
