If I were to have a Deluxe AP, show up on a blockout day and buy the blockout day ticket for $50, could I use the ticket and the $50 cost towards upgrading my Deluxe AP to a Premiere AP?

Yes you could but why? Just upgrade your AP when you arrive and skip buying the blockout pass. Don't forget that the Premiere pass includes parking. If you paid to park then bring your park ticket with you to the ticket booth or guest service window (left of Disneyland gates). They will credit the amount paid towards the AP upgrade.

Yes, you should be able to use that ticket purchase towards the upgrade, but you would have to upgrade by the end of the day on the day you used the ticket...so there is no real advantage to buying the ticket vs upgrading. You can't use the ticket on Monday and decide to upgrade and apply the ticket on Tuesday or later...must be upgraded on day of use.

My friend did this last year when he upgraded from a SoCal Select pass to a regular SoCal pass, but I don't know the details.

If you're planning to upgrade to the Premium anyway that day, I don't know why you'd bother buying the blackout ticket though - it's just an extra step. You can just upgrade from Deluxe to Premium on the blackout day and be done with it. The cost will be exactly the same.

You receive a blackout day exception on the day you upgrade, even if the day you upgrade is blacked out under your new pass.

Even after all my trips through the years, I was not aware of any places outside of the park to upgrade APs, since I never had a need for it before.

Yes you can do that, and as Huff said if you've paid to park you can also apply that $15 to your upgrade. That said you can upgrade to a PAP at the ticket booths if you already have a DAP. Since you'd need to buy the blackout ticket ask at the ticket booth about upgrading to the PAP, I'm sure you can do it there with no problems.
