My friend will be staying at Pop at the end of January. He will be flying into MCO a day early and staying in a hotel close to the airport. My question is will he have to use transportation from the hotel or can he go back to the airport and use the magical express?

Yes, he can go back to the airport and use Magical Express.

Just to make it easy, have him give the Reservations folks a flight number (any flight number) for the day he actually needs to use Magical Express.

Tell him DO NOT use the yellow tags. He will have to claim the bags himself, then take the shuttle to his first night hotel, then the hotel can return him to MCO the next morning, and he checks in and boards DME with his bags. He will need to tip the bus driver and Disney Luggage Services for handling his bags.

Carol's information is correct HOWEVER the OP might consider paying for a car service or cab. Many of the airport hotels schedule the shuttle service back to the airport, generally every 30 minutes. The OP will have to go down to level one, find the DME desk, check in, wait for a bus and then go to their resort.

A cab or car service will get them to their resort in 20-30 minutes. Ask the hotel, they can probably get you a service or cab with a flat rate.
