We are arriving to Orlando airport in the evening and have a room reserved at the airport hotel for that evening, before taking Magical Express to Disney the following morning. Can we utilize DME for a checked bag? Will they just hold ag our resort once they see the tag is for a check-in the following day?

You can only use DME on the day you arrive at your Disney resort, and the day you leave your Disney resort.

PPs are correct. Since you wouldn't be eligible for DME on the day you fly into Orlando, there's no telling what would happen to your bag if you put the yellow tag on it. You should not use the yellow tags and instead collect your luggage from baggage claim to bring your airport hotel. The next morning when you go to DME, the driver will store it under the bus for you and you can leave it will Bell Services at your Disney resort if your room isn't ready yet.

Not strictly true. You cannot use it before the day you check-in, but you can use the MCO-WDW leg after the day your hotel reservation starts. And the WDW-MCO before the day your reservation ends.

Useful if you have people on one reservation but are arriving/leaving on different days.

Good point. Disney resort guests are entitled to one trip FROM the airport and one trip TO the airport at any point during their stay. It doesn't matter when, but it must fall between your dates of stay.

You should be able to have Hyatt Bell Services claim your luggage from the carousel, then deliver it (yellow-tagged by you overnight) back down to DME to be put on the truck. All for a fee. Plus tip, of course.
