would it be much easier to do a tour with greyline tours for either or both?

Have you already priced out a car rental for two days? I think those bus tours tend to run about $60 per person (but please call to confirm because I am not certain). If the car rental price is obscene and more than the price of the two day trips, then just do the bus tours. If, however, the car is a steal, then I'd go on my own. The LA tour, however, is probably more convenient, unless you have a great GPS and/or are familiar with LA. It might be more of a headache than it's worth to do that tour on your own.

It's easy to get to Universal from DL - straight up the 5 to the 101 and off at Universal.

The traffic is the trick - as long as you avoid rush hour you could do it in an hour pretty easily - maybe even 45 min.

Try priceline and costco (if you're a member) for car rentals. I've done well with both.

Monday through Friday heading north from Disneyland after 8:30am usually avoids the rush hour mess. Heading back south can hit traffic until around 7 or 8. But if Universal is open past 8pm should be no problem. Weekends traffic heading south from Universal can have moderate to heavy traffic most of the evening.

Actually between about 7:30 and 9 in the morning is the worst of it and you'll be headed in the main commute direction from DL. Take advantage of car pool lanes - they move quickly. Just don't cross the solid line to enter or exit - it's a hefty ticket. Checking a map there's not much of one for that trip, but it goes for a little way north of DL on the 5.

We drove to Universal Studios last time and found it to be pretty easy and we are from Australia so not used to LA's freeways at all!
