It is before you get on the ride. You give it to the CM you see last. The one who guides you where to sit, etc..

The only one we found misleading was Peter Pan. The wait time was posted as 20 minutes, so we thought why not? It was 45 minutes later when we finally got on. That made us re-think all of the wait times that were listed as 20 minutes. The 20 minute wait time seems to be Disney's default wait time.
