You can definitely buy one-way transfers. We fly in from Chicago and get an MCO-area hotel via Priceline for around $28 to $38. Bid the highest star level and they will have a free shuttle. Then we have Happy Limo pick us up at the hotel. We prefer a towncar, but in theory you could take the free shuttle back to the airport in the morning and then take the DCL bus. Arriving a day ahead really helps reduce the stress.

Barb is great--I'll echo all of the above. If you bid Priceline, you do not want to go below the 3* level at MCO. All 3* hotels have shuttles. If you go below that, you'll spend more on cabs than you will save by going to a lower grade hotel.
For 3 or more people, a towncar is sometimes cheaper than DCL shuttle. The towncar will pick you up at your hotel. Alternatively, you can take the hotel shuttle back to MCO and catch the DCL transport there. Yes, you can but one way DCL transfers. You can purchase them in advance, on the ship, or at the port.
Just to add my two cents, we are arriving the day before our cruise in April. We are getting in pretty late and so I bid on a 3* MCO on Priceline and landed the Crown Plaza. They have a free shuttle that we will take from the airport to the hotel and then back to the airport the followig morning to hop on the disney shuttle.
As an extra, we plan on taking the disney shuttle back to WDW (the Contemporary actually and then probably a cab to WL) following the cruise as we are spending a night at the Wilderness Lodge before returning to reality.
