If I reserved a midsize but used an upgrade coupon that brings me to fullsize do I still get to use the Aisle?

Nope. You will use the fullsize aisle, which will have only fullsize cars in it.

I'd see if the coupon could be used to go from the level down from midsize to bump you up to midsize.
