Do you call to request

Calling the resort will not work, what you need to do is when you book your reservation make your requests at that time. Your requests will then be noted on your reservation.
If you have already booked, then you can call CRO now and put in your request. If you made your reservation through a travel agent, the TA must call to put in the requests.
It appears from reading other posts that the assignment desk at the resorts only gets the reservation somewhere between 3 to 5 days prior to your arrival.
 
You call to request (call Disney's main line and they'll connect you with whomever you need to talk to) and then when you check in say something again. That's your best bet.
 
I only request upper floor when making my reservation.

I purposely check in as early in the am as possible to get the best room location (8am to 9am). When you check in early, you have more rooms to select from. When they show you on the map where your room is, you can say "very nicely" do you have something else? :) And give them your request.

We've always received the best rooms, views, locations by checking in early. The room might not be available early in the morning, but it's ours at 3pm:)
