Not sure on the card, but I took a 16" X 20" sign wishing our godtwins HB and all the characters signed that and posed holding the sign. They were thrilled when they got it and the photos!

They will sign anything as long as it's a "kid friendly" general birthday card No adult cards ect (Not that I'm saying you would bring that ) But they have certain rules the have to follow. But a birthday card, they will sign
If your niece is into Princesses, then I would suggest making reservations at CTR. I had lunch there for my birthday and received a card signed by the Princesses after my meal.

I don't see why they wouldn't. On our last trip, we had breakfast at O'hana for my youngest's b'day and they actually presented him with a signed card when we arrived.
Yes, I imagine that they will sign pretty much anything. I was given a signed birthday card at breakfast at Cape May.
We also were in line at the Princess Gathering with a princess balloon, one of those big ones with pictures of all the princesses. One of the handlers suggested getting the princesses to sign that. So, I think that they will sign anything.
Do you want them to sign it in front of her or be a surprise for her?
We did this on our cruise and it was great. I got it off of Pinterest. We took a large picture frame with an extra large mat around it. I took the mat out of the frame and left it at the Guest Services desk at the beginning of the cruise. They had every character sign it and now I have a nice 8X10 picture of the family in that frame.
I'm sure you could do the same either at your hotel or at a guest services desk somewhere, then it could be a surprise when she opens the card and they've all signed it. Then you also don't run the risk of it being damaged or trying to make sure you find everyone to sign it.

If you're planning on using a normal-sized birthday card, you may also want to bring a clipboard to make it easier for the characters to hold, and give them a solid surface at the same time.

Better yet, if you have a video camera record them sending Birthday Wishe to her. My niece loves Ariel and one year when we went to Norway for dinner I asked Ariel if she could say hello to my niece Gianna whom she met last year at Disney. Ariel was wonderful and I recorder her saying a big hello to my niece and that she remembered meeting her...my niece loved it.
I would just have your vide camera ready and when you go up ask the princess if she could say happy birthday to your niece.
WE have done all kinds of crazy things over the years. Had postcards signed, Made signs and had Characters sign and pose for picture with sign. This past May Pluto sent my BFF ( pluto lover) a big ole lick via pic msg. WE had made a special sign for a friend and wanted Tigger to sign and pose with it. next thing we know we had a Bday card and the sign.
