Yes you can have backpacks in line, but for the coasters the pouch to store your purse/bag is on the smaller side and a full size backpack probably wouldn't fit. Also on the water rides, there is no place to put it to keep it dry. I'm not sure about placing the backpack on the station while you ride the ride, I can't say I recall seeing that at DL (I know I have seen that at Six Flags)

Yes, you can (and should) take them on the rides.
No, you cannot leave them where you board.
Yes, there are lockers.

When I take a backpack, I typically put it on the floor, and put my leg between the straps. I've never had it fly out.

I don't recommend leaving your backpack at the station. There are no shelves to put them on.

As others have said, you'll welcome to have backpacks with you in the line, but you'll have to take them with you on the ride. Most times you can just shove them between your legs on the floor and hold them in place. Most of the Disney coasters are pretty tame so you generally won't have a problem holding onto the pack. I've done this on Big Thunder, Matterhorn, and Space.

It's a little more awkward on Star Tours and Indy because there isn't a lot of space on the floor. For Indy, avoid being seated in the wheel seat (front row) because the steering wheel gets in the way and makes it hard to wedge things in/out.

If you're a large or tall or long-legged person, you might have some problems on Matterhorn. The cars can be pretty cramped to start with, so getting a backpack wedge in front of you, might be uncomfortable.

The only ride where you can leave things at the "station" is Soarin. If you can't fit your bag in the under the seat compartment, you can leave it on the floor. The CMs were directing guests to do that last week, especially with packages and other bulky items. I should add that for this ride, you cannot hold items with you. They either must fit in the seat compartment or be left on the floor. They don't want folks to accidentally drop something and hit one of the riders below.

Friends of our left their backpack while going on a ride. When they came out the CM had changed. They asked and pointed at their bag and it was handed over without a second glance. They quickly learned it is not a CM's job to watch your bag and anyone could have claimed it as their's!!

You can absolutely take your stuff on. Even when things don't fit into the mesh pockets, I just hold on to the backpack/purse. Even on Screamin', etc. Make sure all pockets are shut, of course. :)

Don't leave anything anywhere. First, you'll come back to find it surrounded by bomb-smelling dogs. Second, you probably won't be offloading in the same place you loaded, so you can't even get back to where you started to begin with. Third, a CM that lets you is a little confused. :)

It's not like Universal where there are some rides where you absolutely have to put them in a locker. (I assume U.Hollywood has some of those rides like U.Florida does?)

Beside Grizzly River Rapids there are FREE lockers that you can use for backpacks and anything else you don't want getting wet while enjoying this attraction. We put our shoes in there as well and wore flip flops!

instead of a backpack I took this yoga bag that went across my body. carried just enough in it, and was very thin. so i never even had to take it off on most of the rides. i think Soarin' i put it under the seat, and in jumpin jellyfish we all left our backpacks on the ground. but otherwise, i always kept it on me.

Since my backpack or bag is usually too large to fit in those little mesh pockets on the rides, I put my foot through one backpack strap loop and make sure all my pockets are zipped up. If I plan on going on a lot of rides I rent a locker by the entrance for the day.
