2 transport questions

I am thinking about renting a car at the airport for my trip to WDW for the first 2 days.

1) Can I still use ME for my lugage?

2) Is there a drop off/Pick up for car rentals at the Poly resort?

No, and No. DME isn't designed as a luggage transferral service. If you want your luggage to be transported then you have to be on the bus.
And no, there are no rental companies at the Polynesian. But, the Car Care Center is not too far away. You would make your rental reservation, using the CCC as the rental place, then you call the CCC and they will send a shuttle to your resort to take you to the CCC to pick up your rental. Then, when you are ready to return the car, take it to CCC and they will shuttle you back to the resort.
There is also a car rental desk at the Dolphin. We used that a week or so ago and it worked out quite well.
I would use DME for the trip to the resort from the airport and back again. and then rent a car from one of the on-site areas. I know that both Alamo and National rent at the CCC and the Dolphin.
