We are travelling from MCO (from the UK) to Cocoa Beach the day before our cruise so we are there on time. What would be the best transfer company to use? Any recommendations? Also we are hiring a car when we disembark our cruise at Port Canaveral. Can anyone recommend a car hire company nearby? Thank you.
 
The "best" is sipping champagne in the back of a limo from the airport to the Port!

But since you are coming in a day early, I always suggest to look into one-way car rental first. Then you have wheels in Cocoa Beach to visit the Port restaurants, maybe a grocery store and wine shop too. Folks often pay under $50 per day for a car.

I used A1 Elite Transportation in January and will use them again in March. They are wonderful. We had a limo in Jan but will use a SUV in March. Joanna is the contact that books the reservations. She is wonderful to deal with.

We are taking our first DCL cruise in April and are staying at the MCO Hyatt the night before.
Any advice / tips on using the Disney bus line from MCO to Port Canaveral?
Does the bus get us there in time without rushing to board or being in a huge mass of people?

The first bus out from MCO to port typically departs around 9:15. With subsequent busses departing every 20 minutes or when full, whichever is first. The trip takes about an hour. The port is busiest, generally, from 10:30 to around 12:30. You can decide when you want to catch the bus from MCO to time your arrival at the port.

You just jump into the same lines you would normally use to get the bus to WDW?

Well, the check for DCL transfers is in the same area as the Magical Express check in,

but on the other side of the room.

. The busses for both pull into the same slips,
