We're thinking that maybe we'll rent a car/van to get to the port from OKW next April, one way. Then my DH will take the shuttle to the port after dropping it off.
So can anyone tell me what direction we should take in starting this research and any experiences with families of five?
Also, if we rent from DW, how do the resorts handle this, are the cars dropped off for our use somehow? We also want to rent on the way back to DW after the cruise...can we leave it at the resort we're staying at? How does all this work? But I really need to know one way prices first, if you could. thanks.

Yes, one-way car rentals are available. You can drop of your rental car at a rental car office in Cape Canaveral, and they'll take you to the Disney Terminal in a shuttle van.

But... don't forget about your luggage. You may be able to get a family of five into a small, cheap rental car. However, such cars have small trunks.

The advantage of the DCL bus service or a van service is that they can get your family and your luggage to the Disney Terminal. (You also mentioned renting van; that's a possibility, but it may not be so cheap.)

I believe that the only on-site car rental at WDW is Alamo (the "official" car company") They have some sort of setup that they will pick you up and bring you to their office (near the Floridian, I think.) to get the car. I would think that renting a mini-van would be cheaper than DCL transfers for 5. We have found that it will be much cheaper for us to rent a mid-size than to use the DCL transfers. If you haven't yet, try mousesavers. com, they keep a list of available discounts on rental cars.

I am confident that you should be able to find a rental (van or SUV) that would hold the 5 of you plus luggage - I got 6 and luggage into a mini-van, 5 of us adults, 2 week vacation (read LOTS of luggage or airline limit at least). We dropped off over by Downtown Disney and picked up at the same hotel (used the buses and a little walking). I am thinking it was Avis because they were at the Radisson for that particular cruise.
