I'm guessing your rental car prices will NOT be the same because you're doing a "one-way" rental (Tampa to Orlando) instead of a round trip. Go to a rental car site and check prices each way. Find out how much the rental car will be.
The major rental car companies have locations close to WDW. National and Alamo are at the Dolphin and the Car Care Center, Avis is at the Hilton at DD, Budget is at the Doubletree at DD, and Dollar is at the Wyndham Palace at DD. Hertz is somewhere in the area, too.
Yes, you can do that, but usually "one-way" rentals (returning it to a location other than where you picked it up) are more expensive than round trip rentals (returning it to the same location where you picked it up).
If you book from Orlando-Orlando, what's the cost? Does it eat up all the savings of flying into Tampa?
Another option (and I know some folks like having a rental car) is to not use a rental car, fly into Orlando and utilize Magical Express & Disney transportation.
You can return the car to Budget locations besides the Orlando airport, such as Budget at Downtown Disney. It might cost a bit more than an airport return, but it will save you time.
I'm doing the opposite - I'm flying into MCO and leaving from TPA. For my dates (late Feb - early March), the car rental was actually cheaper than doing MCO round-trip. So, I saved on airfare and on my car rental.
