We're taking I-90/I-77 down to Charleston WV for the first day, then I-77/I-26/I-95 to Jacksonville FL day 2, and into WDW the morning of day 3. Exact reverse on the way home..

We're traveling with DS1 and DD2, and the grandparents, so the shorter drive days and lots of breaks are likely to be critical, rather than driving straight through :)

Can anyone suggest 'must do' or known/preferred break points where the kids can wear off some of their energy along this path?

When our kids were small we stopped at the visitor centers for each state. They could run around a bit and it gave me the opertunity to clean the bugs off the windshield. We also went to the dollar store before we left and made a hugs treat bag. When we stopped they got a new toy to play with.

Any place with a green space for them to run around. The highway-side stops with washrooms are good on a nice day. Just bring a ball or other toys for them to throw around outside and make sure they have a good bit of time enjoying them before packing up.
For your 2yo, I'd recommend a cheap headrest DVD player to watch. We bought a dual set for our DK's pretty decently at Canadian Tire.

DVD players - Check :) Lots of Disney DVDs for the road - Check :)

The 2 (almost 3) year old has a camera to take pics along the way and when we stop on shorter trips, so that seems to keep her happy. We'll see if it'll works for the long trip!

A few people have mentioned balls, etc to throw around in other threads. Will try that out for sure.

Our trip doesn't even take us past South Of The Border since we hit I-95 much further south... Starting to sound like the drive is going to be rather dull :)

Oh the drive is far from dull. The scenery is awesome. We always go this way because of that. DVD tons of movies, cd's the whole family loves to listen to. Toy bag, Small cooler close at hand for drinks and healthy snacks. New teddy bears for them to play with. When there older video games, mp3 players and cameras are awesome too. I like the idea of a ball to kick around. Thats one I will add to our list for sure.
