Can you choose DME times independent of flight times?

If we're flying into Orlando a day before our resort reservations, can we contact DME to let them know when we'd like to use the transportation the next day?
I realize we'd have to get our bags to the bus but is it possible to make DME "reservations"?

Yes - just call DME directly and make your reservation.

I just listed the first incoming flight into MCO the morning after my actual arrival (we flew in late the night before and stayed at the Hyatt) as my arrival flight when I set DME up online because I hate talking to people on the phone if I don't have to. DME doesn't check flight manifests and that seemed like the easiest route to me.

I have also simply called to give alternate times and days. I had members of my party on different arrival/departure timings so I just walked them through the two groups.

Just make sure you get your return to the airport exactly right, as they schedule those buses down to the seat and somebody showing up late is usually denied boarding,
