I'm trying to research the difference between these 2 room categories, and I can't find a layout to see what the difference is. The ones on the DVC website look identical between the 2BR and the 2 BR lockoff. I think a lockoff is a studio and a 1 BR combined, so there is a door between the two that you can access. Is this true?

Correct.

With a 2-bedroom lock-off, the studio becomes the 2nd bedroom and will have a queen bed and a sleeper sofa (except for Old Key West which will have 2 queen beds).

A dedicated 2 bedroom villa will have only one entry door to the hallway. It will not have the small kitchenette that a lockoff has. It should have two queen beds in the bedroom rather than one queen and a sleeper sofa.
The 2 bedroom lockoff will have two entry doors, one for the villa and one for the studio/second bedroom. It will also have the kitchenette and the sofa bed (except OKW). There will be a door to access the villa which can be closed to lock off the studio/2nd br.

@Wood Nymph, can you confirm that a dedicated 2 BR will only sleep 8 people and a lockoff will sleep 9? Based on what I have researched and what you have said above, that is the only thing that makes sense to me. Because the studio would have the murphy bed, a dedicated 2 BR would not have the murphy bed, correct?

That is true. The lockoff will have the fold down Murphy bed (for kids or shorter adults) in the studio portion. It is not in the second bedroom of a dedicated two bedroom.
