Right now we are booked in a Lion King room for our upcoming trip. Everyone really wants to stay in the Finding Nemo room. Is this something that can be requested when i check in online?

No. You will have to call and see if there's any available Finding Nemo Suites. If there are ask if you can switch. You may have to pay the switch fee.
