We are staying onsite Universal for the first time. Do I have to purchase park tickets through Universal to still get the front of the line/express pass? Or can I use my Work advantage and purchase discount park to park tickets?

You can buy tix anyway you want to. Your tickets aren't connected to express pass in any way.
