I would definitely opt for the electronic pass on the rental. That way, you just go right through and do not have to stop. In fact, you don't even slow down because they have totally separate lanes.

The rental company usually charges a daily fee for having the toll pass so it won't save $ in the end but it will save time.

How much time are we talking about? I'm no expert but I have been traveling in florida on these roads during spring break for a couple decades, I don't think I've ever been in a line more than a couple cars long.

I fairly routinely see queues of 10 cars at the cash change/receipt lanes on the Beachline. In reality that is probably not more than 5 minutes but it can SEEM like half an hour while you watch cars zip by without slowing.

The referenced SUNPASS page for Rental cars SHOULD be viewed because it has very useful information about the WIDE RANGE of fees the different rental companies charge to use electronic tolling.....

There is seldom a back up in the exact change lanes and one roll of quarters will suffice for a round trip.

as OP said, please check the fees involved with using the rental car transponder. They charge a fee for EVERY day you have the car if you use the transponder only once!. So if you do a 1 day rental (pick up MCO, drop off at port, same day or next AM), no big deal but if you have the car for a week and use the transponder once, they charge about 2.50 a day for services not rendered.
I try to stay away from these things if I have the option (or knowledge). I have been burned by this in the past.
On the other hand, using the cash lanes (exact change is a good idea) but I want a paper receipt so if I am charged for electronic tolls, I can prove I paid cash. the lines at the toll booths can get a bit long but build that into your travel time.

Why would anyone use the rental car's transponder? I have seen companies that charge $8 per day. For $5 you can buy a sticker from Sunpass. Sure it only works once but it is cheaper than using the rental car's transponders.

Sitting behind a line of cars where drivers are searching for quarters seems like a complete waste of time to me.

Just bring a roll of quarters with you. That's what we did. They were easy to carry and we always had exact change. The fees on the electronic box were ridiculous. It would have cost us double what we paid in tolls.
