Budget does not charge a one way fee. It's easiest to have only the driver take the shuttle to the rental office, which is off the port. Why struggle with luggage and such twice?
Be sure to sign up for Budget's free loyalty program. You skip the line at the rental office! I saved about thirty minutes in the Fall of 2012.
Budget is located on airport property for a quick departure.
Use the Gas Buddy app to find cheap gas on the way. Remember to bring some change for the tolls, $2 total. To use cash on the Beachline (Florida 528) keep to the right for the cash only lanes, kind of like an exit ramp.

I was checking all the major rental agencies a few days ago for one way rentals for our next cruise and only Thrifty quoted a $50 drop off fee.
