I will probably be making my reservations tonight for our first time ever to Universal. (been to orlando six times - always gone to Disney)
I'm trying to decide if I should book through Universal Studios Vacations, or book everything separately.
Portofino Bay has rooms available for the dates I want. If I purchase my park tickets online, separate from my room reservation, will I still be able to use my room key for Express/ FOTL?

Yes you can! Just show 'em the key at the rides.

As nativetexan has pointed out, yes you just show your room keys for FOTL access.
Have you looked into the 5 for 2 Bonus Pass available only online?
It is not part of a package, however, we always purchase them and make room only reservations.
You get 5 consecutive days of both parks (hopping allowed) from the first day you use it. They cost about $95-$100 each. ::yes::

And yes, WDW, the 5 for 2 is the main reason I was planning on booking everything separately.

And if you buy the bonus passes online using a Univeral Mastercard, you can get 5 for the price of 4, so you can save even more.

Since there are only three of us, the 5 days for the price of 2 works out better for us.
We'll arrive late on a Sunday and if we feel like it, we can pick up our tix and check out a couple of things before we really hit the parks on Monday and Tuesday. Then, we'll still have two days left on the tix to use on a whim. We'll be in Orlando for about 5 days and we were hoping to catch SeaWorld as well.
But hopefully the MasterCard info can be of use to others looking for tips! Thanks for posting. :hyper:
