A good place to start would be with a guide book such as Simon Veness' Brit's Guide to Orlando. All good book stores will stock it. It will answer all of your questions, and a whole lot more. To respond to some of your questions:
Transport - it is perfectly possible to get around on public transport, but if you can hire a car for your holiday I think it would help make the most of your time.
Children - we've never been to WDW with children but despite (because?) this we've had a brilliant time on each trip. Anyone who says that Disney/Universal is only for children has never been to Disney/Universal!
Characters - pop up frequently, so no shortage of meet n'greet opportunities.

Hi rings
Welcome to the DIS :bounce:
Wow - your first time how exciting :) Is there just two of you going?
Have a look at the Ticket Page (http://www.wdwinfo.com/wdwinfo/tickets.htm) here on The DIS to see what type of tickets are available. There are loads of opportunity to meet all the different characters especially Mickey and Minnie. You can even have Meals with different characters.
The DIS is a wonderful sorce of info, any questions post away. You will find it best to have one topic per post, this will help you get all the advice you need. :)
On Sunday's at 7pm we have a 'Chat' with different topic's - try and pop in for that.
I definately agree with getting Simon's book.
