HI,
We will be checking in with our AP discount on the 19th with our AP voucher. When my sister arrives on the 20th, can I use my AP to get the AP discount on her room?

Yes, I've gotten an AP discount on more than one room.

Policy states that the person with the AP must be staying in the room and can only be used for one room. I've heard others say that they have done it but be prepared if they ask for proof of two. Might want to ask when making ressie just to be safe.

You use to be able to get up to 3 rooms with one AP. But Disney has changed the policy so now it is one room per AP. If someone else in your family has an AP you could have them listed on the second room and use their AP for the discount for your sister.
