We have reserved two rooms at a value resort for my family. We are purchasing the dining plan for everyone. My room will have my husband, our daughter, our son's girlfriend and me. The other room will have my 22 and 17 year old sons. Since my son and his girlfriend may be eating separately from us, do I need to list them as staying in the same room so they don't have to use two separate room cards when ordering a meal for the two of them? Or can we combine all meals for both rooms together?

That would be, by far, the simpliest solution, but not just for that reason. I would book your son and his girlfriend in one room and your other son with you. The reason for this suggestion is DDP credits are assigned per room, not per guest.
In reality, each guest has access to ALL of the meal credits for the room. That could really complicate matters -- not only when one son is dining with his girlfriend, but especially for CS meals and snacks. If you list the couple in the same room, then they are responsible for their own food, and they can't take anyone else's -- or have the other son eat all their food.

I thought that might me the best. While I trust my son, I believe it will be difficult to ensure everyone is getting their fair share if we are not eating all of the meals together. Adding to the problem, now my brother's family is thinking of going at the same time and he asked if they can pay and put one of their kids in my extra room (because they have 5 people, too). That means one of our rooms will have a kids DDP attached to it!!!!!!!!!!! I think the only way this will work if they are going is to insist everyone eats all our meals together. This may take more work than I am willing to do...............
