We travel to disney at the start of the the school summer holidays each year (DW is a school teacher, we have no kids as yet), most years we only go the once but had a second trip this year. What I am wanting to ask is as we would only get 1 use out of annual passes it seems to be cheaper to buy the 21 day UK passes rather than an annual premium pass even with the DVC discount. Is it possible to get the 21 day pass upgraded to non expiry so we could use it on a second trip the following year?

I think your only option would be to purchase actual non-expiration tix, which is not such a great deal compared with the 21 day passes from the UK Disney store if you are only staying for one three week break. An annual pass should pay for itself even although you are only visiting once per year, especially with the food/shopping discounts!

How often do you use the extras from the premium pass like water parks, Disneyquest and Pleasure Island? I worked out that I needed to use them at least 4 times to make it worth getting the premium pass instead of the ordinary. I then realised that $100 off the ordinary annual pass became the best possible value for me - about £175 and covering two 3 week trips.

How about staggering your holidays. If you have a 3 week vacation every year buy a DVC AP and go three and a bit weeks later than you normally would. You can use your AP on this trip then go at the start of the school holiday the following year and your AP would still be valid so you get 2 trips from one AP making it the best value.
